摘要:采用高效液相色谱法(HPLC)建立发酵液中甜菜碱含量检测的新方法。使用 ZORBAX 300-SCX(5 μm,4.6 mm×250 mm)阳离子色谱柱,以乙腈:0.05 mol/L磷酸二氢钾比例为15∶85(V∶V)作为流动相,检测波长195 nm,流速1.0 mL/min,柱温30 ℃,紫外检测器检测。甜菜碱在0.005~5.0 g/L范围内线性关系良好,相关系数为0.999 9,平均回收率为103.3%,方法定量限为0.025 g/L。此方法具有样品前处理简单、检测灵敏度高、分析时间短、回收率高、重现性好等优点,可适用于发酵液中甜菜碱含量检测。

Determination of Betaine in Fermentation Liquid by High Performance Liquid Chromatography

Abstract:A method for the determination of betaine in fermentation liquid is established by using high performance liquid chromatography (HPLC). A cation column of ZORBAX 300-SCX (5 μm, 4.6 mm×250 mm) was used with Acetonitrile: 0.05 mol/L Dihydrogen phosphate 15∶85 (V∶V) as mobile phase at flow rate was 1.0 mL/min, detection wavelength was 195 nm, the column temperature was 30 ℃ and UV detector. Betaine had a good linear relationship in the range of 0.005~5.0 g/L and the correlation coefficient was 0.999 9, the average recovery was 103.3%, the detection limit was 0.025 g/L. The proposed method has the simple sample treatment, low toxicity, high detection sensitivity, short analysis time, high recovery rate and good reproducibility, can be used for the detection of betaine in fermentation liquid.
